概述
概述
细胞因子( cytokines)是由免疫细胞(淋巴细胞、单核巨噬细胞等)及其相关细胞(血管内皮细胞、成纤维细胞等)合成和分泌的调节其他免疫细胞或靶细胞功能的生物活性物质,属小分子多肽或糖蛋白。细胞因子种类繁多,生物学活性广泛,作用机制各异。细胞因子作为分子信使,允许免疫系统细胞彼此通信,以产生对靶抗原的协调,在许多疾病中具有调节和效应功能,因此细胞因子及其受体可用于免疫治疗。
细胞因子分类
1.按其细胞来源,细胞因子分为淋巴细胞产生的淋巴因子( lymphokine,包括IL-2、IL-3、IL4、IL-5、IL6、IL-9、IL-10、I-12、I-13、I-14、IFN-γ、TNF-β、gm-csf等)、单核-巨噬细胞产生的单核因子( monokine,包括IL-1、ⅡL6、IL-8、TNF-α、G-CSF、M-CSF等)和其他细胞(上皮细胞、血管内皮细胞、成纤维细胞等)产生的细胞因子(如EPO、IL-7、IL-11、SCF、I8、IFN-β等),但不包括免疫球蛋白、补体及一般的生理性细胞产物。
2.按其主要功能,细胞因子分为白细胞介素( interleukin,IL)、干扰素( interferon,IFN)、肿瘤坏死因子( tumor necrosis factor,TNF)、集落刺激因子( colony stimulating factor,CSF)、转化生长因子β( transforming growth factor-B,TGF-B)、趋化因子家族( chemokine family)和其他细胞因子。
特点
1.低分子量的分泌型蛋白质,分子量<80 0000;
2.产生具有多元性,即单刺激可使同一细胞分泌多种细胞因子,一种细胞因子可由多种细胞产生,并作用于多种靶细胞;
3.正常静息状态细胞极少储存,需经激活后合成分泌;
4.生物学效应极强,导致细胞行为的改变;
5.以非特异方式发挥作用;
6.大多通过自分泌或旁分泌方式短暂地产生并在局部发挥作用;
7.需与靶细胞上的高亲和性受体特异结合发挥生物学效应;
8.构成细胞因子网络相互诱生、调节和影响。
抗肿瘤机制
(1)控制癌细胞的生长和促进分化;
(2)调节宿主的免疫应答;
(3)对肿瘤细胞的直接毒性作用;
(4)破坏肿瘤细胞血管和营养供应;
(5)刺激造血功能,促进骨髓恢复。
分型
1.激活免疫疗法
在激活免疫疗法中,如粒细胞集落刺激因子(G-CSF)可以刺激外周血干细胞(从病人血液中提取)产生淋巴细胞,经过体外与肿瘤抗原共培养后,再输回到病人体内,并结合刺激性的细胞因子以增强免疫效应,这样的细胞就可以摧毁携带相同抗原的肿瘤细胞,从而达到治疗的效果。白细胞介素-2可以与抗CD3和同种异体反应性细胞融合,生成过继T细胞。这种细胞可以转移到患者体内,可以进一步提高IL-2的抗癌活性。白细胞介素-7和白细胞介素-2可以用来恢复免疫功能缺陷患者的免疫系统,这一研究已经进如临床试验阶段。
2.抑制免疫疗法
在抑制免疫疗法中,主要抑制自体免疫疾病中的异常免疫反应,或者降低正常免疫反应以阻止细胞或者器官移植中的排斥反应。如免疫抑制药物、免疫耐受、过敏治疗等。

Innate lymphoid cells (ILCs) are largely tissue resident and respond rapidly toward the environmental signals from surrounding tissues and other immune cells. The pleiotropic function of ILCs in diverse contexts underpins its importance in the innate arm of immune system in human health and disease. ILCs derive from common lymphoid progenitors but lack adaptive antigen receptors and functionally act as the innate counterpart to T-cell subsets. The classification of different subtypes is based on their distinct transcription factor requirement for development as well as signature cytokines that they produce. The discovery and subsequent characterization of ILCs over the past decade have mainly focused on the regulation of inflammation, tissue remodeling, and homeostasis, whereas the understanding of the multiple roles and mechanisms of ILCs in cancer is still limited. Emerging evidence of the potent immunomodulatory properties of ILCs in early host defense signifies a major advance in the use of ILCs as promising targets in cancer immunotherapy.
