Neuropeptide Y (NPY) is a very abundant neuropeptide in the brain and was first isolated from porcine brain in 1982. It belongs to the so-called gastrointestinal-brain peptides and is evolutionary highly conserved. Owing to its strong appetite stimulating effect, NPY is considered as the most potent orexigenic neuropeptide. Moreover, it is also involved in several regulatory cycles, e.g. concerning the cardiovascular and immune system, bone homeostasis, sleep, stress response and anxiety. These physiological functions explain its implication in human diseases like obesity, hypertension and arthrosclerosis, stress and mood disorders as well as cancer. NPY mediates its pleiotropic functions by interacting with four different G protein-coupled receptor (GPCR) subtypes, the human Y receptors (hYRs), with the hY1 R subtype being most frequently implicated in all of the mentioned physiological and pathophysiological processes. To understand the diverse (patho-)physiology of the NPY/hY1 R signaling, hY1 R-preferring NPY analogs constitute important pharmacological tools.
