Progranulin (PGRN) is a cysteine-rich protein that is crucial in diverse biological processes, such as cell proliferation and embryonic development and is known to perform various biological functions, such as protection against vascular disorders and neuronal disorders. PGRN can be subjected to protease-mediated cleavage by enzymes, such as neutrophil elastase to release granulin (GRN). Both PGRN and GRN have biological activities, although often with those opposing functions in immunoreactions. In a mouse model of rheumatoid arthritis, PGRN was found to inhibit inflammation by suppressing the TNF-α-mediated intracellular signaling pathway. Also, PGRN is a neurotrophic factor that promotes cellular survival and stimulates neurite outgrowth in the nervous system.
